DMSO concentration - (Jan/26/2008 )

Hi - what general DMSO concentration is good to use? My cells from ATCC say to use 5% DMSO (v/v). Some other people just use 10% DMSO as a standard.

Also, I have read on the freezing/thawing post that this low DMSO will not be toxic to the cells, but people I've worked with tell me DMSO is toxic*toxic to cells. I'm wondering what is your opinion on this, because I feel I have to rush every time I freeze cells, and I end up accidentally knocking some stuff over (luckily not the cells!).

protocols depend on experience; we have cells we freeze with 5% DMSO and others with 10%; if the data sheet of a specific cell of ATCC recommend a freezing protocol, it should work; if there is no experinece one has to run an optimazation protocol

we use 10% DMSO for storage of cells. I don`t think that those 5 minutes where the cells are in DMSO, can be so toxic although I also rush every time blush.gif . Anyway, every time you thaw cells there will be some which die but you keep them nicely until recover.
