QuickChange II Site-Directed Mutagenesis Kit Stratagene - (Jan/24/2008 )

I'm going to do site directed mutagenesis for the first time and I'll use this kit.
Like always the product description seems really easy and straightforward, but we know it is never like this.
Can people that have already used it share their experience with me?
Mostly about problems that have arisen and possible tips!

For one aminoacid mutations, you don't need any special tip.
Follow provider recommendations.

This is really a realy nice kit. Honestly, I think it is one of the few examples out there, where a kit is as efficient and easy as the provider promises.

No, I promise. Just a poor Ph.D student who - after having cloned for years with varying success -was happy to find an efficient method. We even used the same technique for cloning PCR fragments (1300 bp). Just think of your PCR fragment as two extremely long primers then you get the idea.

Efficient method, but you don't need the kit!

Buy components separately (from different vendors, though stratagene's pfu-variants are among the best High Fidelity polymerases in my hands) and make your own competent cells. Saves you a lot of money and works equally well.

we follow the protocol that comes with the kit quite faithfully. We use their program to design primers and buy their PFu enzyme. Everything else is bought elsewhere to keep costs low. It works well.

I've bought the kit already. but thanks for the suggestion I'll try to do it when I'll end the kit.
