Linker Sequence - Which sequence between 2 functional domains? (Jan/21/2008 )

Hi,

I have an expression vector coding for a protein with 3 functional domains. I want to delete the middle domain but doesn't want to disturb the flexibility of the other two domains. So, my question is which sequence is commonly used for this purpose to put in the middle?

One that is frequently used is a repeat of the sequence GGGGS 3-4 times. You might want to avoid repeats in the nucleotide sequence to reduce recombination effects.

Adding glycines is a good idea if you need some flexibility.
