XTT - cell viability (Jul/18/2004 )

I have a very simple question? when you prformed XTT at the end of the incubation how do you take the readings. The problem is in this method cells remain alive. So should i take the reading in the same plate in which cells are there or should i transfer the media in another plate. While transfering this if i mix the soup then my cells are detaching from the plate and come out in the media. Will this interfere with my absorbance.

If you want to keep your cell cultured after XTT assay, simply read the plate with the cell in place. The reading won't kill your cells. You can keep the cover in place to prevent contamination.
