SYBR green ready mixes - need opinions please (Jan/17/2008 )

Hi guys, we've just got a new ABI 7500 and I'm trying to decide which SYBR green mix to buy, as they're all really pricey.

Just foung a pretty cheap one (comparing) from Sigma, SYBR® Green JumpStart™ Taq ReadyMix™, and just wondered if any of you have experience with it.

Any other advice and/or comments will be more than welcomed.

We use a "home made" mix with an ABI 7500 and it works really well, because as you said, commercial mix are really pricy.

I've been trying a home made one, but isn't working. Any tips? please... what Taq are you using? as i think my problem might be there. I've just been using Invitrogen Taq polymerase, with CES as enhancer (a mixture of BSA, DMSO, DTT and Betaine), and MgCl2. I've tried 0.5x, 1x and 2x SYBR green I, with 1 or 3 mM MgCl2 and although my reactions worked (as seen on gel) I dont get any fluorescent signal huh.gif , and my melting curves are just a joke sad.gif

Which reference dye you added in your home made mix? ROX or FAM or VIC? Where did you get them? Please give us your comments. Thanks.
