Mouse embryo sections - how to see optic stalk? - (Jan/04/2008 )
Dear all,
I am trying to make some sections with mouse embryos between E10.5 to E14.5. The embryos are X-Gal stained (wholemount) and then paraffin-embedded. I want to see the optic stalk and the eye together, I tried both cross sections and sagittal sections but so far I couldn't succeed to see any optic stalk at all, the eye part is OK. Does anyone have any suggestions for the embedding and sectioning steps?

I just had a quick look on google and I saw some nice pictures of optic stalk and the developing eye using transverse sections. Not sure if this helps!
How thick are your serial sections?

yes, i saw lots of nice eye sections with optic stalk! i'm making sections at 7, 8, 9 microns. it's really frustrating.

The sections I saw online had 1 micron thickness....!!!
