Cross reactive epitopes - Cross reactivity (Dec/14/2007 )
Hello,
I work on viruses and antibodies raised against these in the human serum sample.
I work with 3 viruses in the same family, I found that the serum samples are positive to all the 3 viruses, which should not be strange, but I would like to see if there is any cross reactivity among the antibodies, say like if antibodies against 1 of the viruses is the reason for it to give positive results while using another virus.
I do the ELISA by coating then plates with the virus overnight, block it for 2hrs and then serum sample. detection is by HRP.
I have gone through the literature about the virus and was not able to see anyone mentioning about cross reactivity, but I do need to prove by ELISA or anyother method.
I have heard of Epitope Blocking ELISA, but I am not sure it is applicable here as I am using 3 different viruses.

Many epitopes are derived from viruses or bacteria, so they could interfere if its from the same family or their relative. It depends on the region that the antibody recognizes.

the antibody recognises the capsid of the virus and hence the problem, as i have found them cross reacting, is there any way to prove it.

Could you look for antibody reactivity for each virus separately with each antibody. Also if the recognition sequence for the antibody known or can be found out, then its simple. Also if you have the viral genome sequence, might be helpful to compare sequences. But otherwise, I am not sure.
