Also, has anyone used N-Ethylmaleimide to determine thiol concentration. If you have can you help with a protocol. I have attempted to use it, but I never get a high enough color (I add sample, add equal volume of 0.375M NEM, incubate and add NaOH). Is this correct? Or if you know of another really good method please let me know.

Have you considered using DTNB (Ellman's reagent). It's very easy!
