Infection of human cell lines grown in suspension? - Infection protocols (Nov/06/2007 )
I am currently studying the effects of bacterial infection on macrophages. I have a macrophage cell line that grows as a suspension. Does anyone have any reputable references in journals that I can look up where the protocol for infecting suspension cell lines is detailed? I have tried searching on the internet, but I keep getting refered back and forth between journals.

my labmate is doing similar experiment as yours.
She is infecting a human pathogenic bacteria into human macrophage U937. This cell line is suspension. But she induced it to become monolayer using certain chemical before infection. 2-3 days before infection, she count the cell number she need, induced in a flask, 6-well plate, wait for it to attach and change media before start infection.
It is easier to do infection with monolayer, easier to handle especially on washing steps to eliminate extracellular bact after infection. just rinse few times with HBSS or whichever buffer you used for your cell. No centrifugation is required.
Hi, I always infect suspension cells using a 'spin infection' protocol. Let me know if you need it, my method is adapted from a paper (which I can't remember what it's called!)
