TRANSFECTION IN HEPG2 - (Nov/01/2007 )

HI Guys,

I am working with gene regulation in fish ovary, I am checking the effect of a particular tarnscription factor on my gene of interest. to chk this I am planing to transfect them into mammal cell line , We have HEPG 2 cell line available in my lab .

The question is : is it ok if I transfect my genes in cell lines which are non fish in origin ? if yes is it ok to use HEPG2 to show the gene regulation which is originally happening in fish ovary? is it a constraint to use ovarian cell lines for ovarian work? is it publishable and acceptable if i use HEPG2 .

please guys reply, if anyone know the answer.

If the transcription factor has something to do with egg development or regulated by sex dependent/specific mechanism, then I would say it is a bad model to work with.

If you want to check if it functions in a ubiquitous fashion, then yes. HepG2 is not very transfectable, but you can get 30% rate with Lipofectamine 2000.

alright ,can you please also suggest some lines which can be used in this case and which have better transfection rates with lipofectamin 2000.

If you wish to study a particular transcription factor, then use any cell line and it should fine. If its a fish gene and you are using another cell line, I am not sure how comparable they can be. better ask your PI about the relevance of using the HEPG2 cells.

Thanks a lot for your suggestions.
I inquired around, I got one ovarian cell line CAOV ,is it a good and easly transfected cell line compared to hepg2.
