Staining polysaccharides in SDS-PAGE - (Oct/31/2007 )

Hi~

I degraded the polysaccharides extracted from plants.
I want to use the SDS-PAGE to observe the degraded pattern
How to stain the polysaccharides in SDS-PAGE??

unless you are running mekc (micellar electrokinetic chromatography) i don't see how you will separate polysaccharides on sds-page?

maybe mekc on capillary electrophoresis? in which case you can detect with uv at 232 (or 234?)nm.

polysaccharides contain proteins; are you interested in them? polysaccharides are less distinct than proteins and therefore are more difficult to separate

Some has carboxyl groups as well. You can try "Stain-All" dye or use PAS reaction. Do a search on Anal. Biochem for articles on this topics.
