mutagenesis of 6 bases - what is the best option? (Oct/07/2007 )

Hi all,

Does anyone know what is the best way to generate 6 mutations in one cDNA. conventional PCR mutagenesis or quickchange seems to take a lot of time. Any suggestion?

You mean a cDNA that is in a plasmid / vector? or are you looking to clone into a chromosome? or are you just looking to mutate a cDNA directly in vitro for some in vitro experiment? Specific mutations or random?
