How to stimulate monocytes? - (Oct/03/2007 )
Hi,
Does anyone have an experience with cell culture stimulation? I need to stimulate monocytes (CD14+ cells) but the amount of stimulators offered by wide variety of companies, as well as recommended protocols almost scares me… If you could recommend some mitogen that works well for you together with an appropriate protocol I will be more than grateful. I have read about Conc A, PHA, PMA, LPS… - is there any difference in their usage/specifity/quality?

I will say your choice depends on what outcome you are expecting. so my question is, what do you want to stimulate monocytes for?

I'm afraid you need to be a bit more precise. Which proteins? also, how are you planning on measuring this increase?
What population are you interested in selecting for?
Or is it an overall culture stimulation?
Also, how long are you going/need to keep the cells in culture?

I'll try to resume my problem:
I need monocytes to increase their production oof prolactin. The increase will be measured by FACS (in cells) and ELISA (in media). I do not need to culture the cells any longer than necessory for increasing their activity. I do not know if it is better to separate monocytes from PBMC first and stimulate only separated CD14 cell population or to stimulate the whole PBMC culture a monocyte separation perform subsequently.
