About how to find out the gene(s) - (Oct/01/2007 )

I want to know if someone got the genetic disease such as tay-sachs disease, then what is the methods that can be used to find out hte gene responsilbe for the disorder??

perhaps it would be a good thing to post what you have thought of so far? that's a very open-ended question - also, on this part of the forum, we will help you find the answer but we don't usually just give it to you

one thing to consider - what is different about people with tay sachs? do they over/under porduce something? is it a metabolic imbalance? how could you use that information to find a genetic component to the disease?

Thanks! I know the affected people show the loss of particular protein, therefore it cannot undergo the normal metabolsim inside the body. I know the present of protein, but how can i use this information to find out which gene is related to this disease?

Is it:
get the protein --> get the amino acid sequence --> get the DNA sequence from the database?

Well done, a good start. What if no-one has ever entered the gene in the databases, or even looked at what protein is deficient? How could you find out what protein or gene is responsible?

you are right, i feel confused how did the people do to find out the affected gene if that is a first-discovered diease?

Anyone can help??

Look at what is affected (the phenotype, what does it show?) and a family tree helps - is it x-linked or not etc

yes, u might right, however, the victim may show many differences (phenotype) comparing to those normal people. If only one gene is mutated, how can I find out this gene??

can we take some samples from the patient, and then do the chromsome anaylsis, compare the samples between the victim and healthy person, isolate the affected chromsome ......
