effeciency of recombinants - (Nov/02/2001 )

It would help if you told us a little more about how you are doing your ligation. ie. are they blunt ended or directional, how are you blunting or adding adapters to your insert (TA cloning, restriction linkers, or restriction sites built into primers, etc.),are they done with a kit or without. If they are done with a kit, which kit is it? The more information you give, the easier it is to target the problem.

My recombinant clones are very less, eg. 1in 100? can any body suggest me that what is the reason for this?when ever i put vector control alone, there will be less colnies. If i introduce insert large colonies, but recombinants are very low may be 2in 100.Please any body can suggest me that how can improve my recombinants.Thanks in advance.
