Nuclear staining for 405 nm - (Sep/13/2007 )

Has anyone an idea which nuclear stain is to use for 405 nm (fixed or live cells)?

-The Bearer-

Hoechst 33258 ext 366 nm. em 410nm. 450 nm rather. DAPI peaks at ~ 440 nm, but you can pick it up ~ 25% of max at 405 nm.

Always get good nuclear staining with Dapi.

ok, thanks genehunter-1 and rhombus;

we have got a new UV-LASER (405 nm) for CLSM but I get no signal with DAPI; so I think DAPI may not be useful for 405 nm, but I am not sure if the new LASER is really well installed unsure.gif

For fixed cells we use Vectashield +Dapi....this however can go off, particularly if it is left out on the bench. If it is an old batch of Dapi, try a new one just in case.
