Wright-Giemsa staining and phagocytosis assay! - (Aug/30/2007 )
Hi everyone,
Now I'm trying to perform the phagocytosis assay for alveolar macrophages. The target cells are apoptotic thymocytes. To observe the result, my PI recommend to perform Wright-Giemsa staining (according to many references). I try to find out the microscope image of phagocytotic index as well as the phatocytic macrophages from other papers. However, just macrophages eating the particles (bacteria or other particles) were found. It's very difficult for me to imagine the expected images, because to my knowledge, the diameters of macrophage and thymocyte are ~13um and ~6um, respectively.
Hence, could you please help me with the protocol for Wright-Giemsa staining of monolayer cells (in 24-well plate), and the image of phagocytic macrophages?

I am also interesting to phagocytosis assay for macrophage but in my case it's for study bacterial incorporation and not apoptotic cell engulfment. Do you know a protocol or a reference to illustrate this kind of phagocytosis???

Hi,
You can easily find our several articles about this phenomenon. With key words : apoptotic thymocytes and macrophage and phagocytosis (or clearance of apoptotic cells).
Actually, when I try to search other papers, just 1 article shown the microscope image of phagocytic macrophages. So it's a little bit difficult for me when starting this experiment.
