how does lysis solution - (Aug/22/2007 )

hi, guy, during DNA purification ,after adding lysis solution, i see white stuff, in the protocol, it said i have to mix the solution immediately after adding lysis, i do not understand why, thanks!

You just want to make sure you have lysed all of the bugs. Once a few cells lyse, the DNA takes up a large amount of space, which makes the lysis of other cells less efficient. Rapid mixing helps the process go to completion efficiently.
Similarly, when you add the next solution, you want to get that through the lysate quickly, in order to neutralise the base. However, this time, you don't want to be too violent, or you cause too much genomic DNA to shear, which makes the task of getting clean plasmid DNA more difficult.

in both cases, after add lisis and neutralizing solution, it has to be quickly but soft invertions.
