pervanadate - how to prepare pervanadate (Aug/21/2007 )

Hi everybody!

I'm a little bit confused with the preparation of pervanadate by mixing H2O2 and sodium orthovanadate. Some protocols mix equimolar concentrations of both reagents whereas other add an excess of H2O2 and let the tube stay at RT temperature (others say at 4°C). Some people also use catalase after the reaction to get rid of the remaining H2O2.
Has anybody experience with different protocols and can tell me which one gives the best results?

I've only worked with sodium orthovanadate... would that work for you? I'm assuming you need it as a tyrosine phosphatase inhibitor.

If so, Upstate gives a protocol for properly activating sodium orthovanadate by heating and adjusting the pH. I don't know what the policy is for providing commercial links... just google " Sodium Orthovanadate Activation upstate " and it is the first link.

You are rigth, I am using the pervanadate as a phosphatase inhibitor in cell culture.

Do you know if there is any difference between both treatments (pervanadate, or activated sodium orthovanadate)?

Not sure if there is a difference between pervanadate and activated orthovanadate. I tried that activation protocol once and after all that work I decided it would be much more efficient to just buy pre-made 10X cell lysis buffer from Cell Signaling. It did not cost that much and works like a charm.
