why need culture our cells in condition 5% CO2 ?
why need tat condition ?

bcuz most medium was buffered with eagle's salt, which ph is about 7 when balanced with 5% co2.
of course, you can use other buffer systems which required different co2 concentration. however, that is not a common way, so you may need to prepare medium by yourself.
that takes time and error probing...
