Semen testing in ELISA - (Aug/03/2007 )

Is there any reason why this shouldn't work?

um...testing for what? you could put semen in a plate and run an elisa, but what are you testing for?

you can elisa for anything if only the antibodies exist and you are patient enough to optimize

you need proper diluents (solvents) for the access of semen proteins to the antibodies.

I'm trying to detect something that I've already got an assay for detecting in serum... Was just wondering if you can work up an assay to detect the same protein in semen. What kind of solvents could you use? I've only used phosphate buffered saline with tween in it so far... Would that work?

hi,
i personally donot have experience with ELISA on semen. Did u manage to get clear solution in PBS?

i m not sure about which buffer to use but certainly u can bring the final pH to 7.4! as semen has basic pH.

if you already have working assay with you, given case you have recombinant protein for the assay
you can spike the recombinant protein into semen-PBS solution and see still your assay is working!
if you can still detect the protein of interest then you can go ahead with screening.
