Sonicating with Misonix Sonicator 3000 cuphorn - Questions about cuphorn with Misonix Sonoicator 3000 (Jul/30/2007 )
I recently started trying to use the cuphorn for the Misonix Sonicator 3000 and have had trouble with reproduceability in my shearing patterns.
One potential cause of this is that I use the tube holder that came with the cuphorn and it does not fit snugly into the cup. So when I turn on the machine to start sonicating, the tube holder bounces around. I don't see it moving up and down vertically, it more just moves in circles around the cup. I am very unsure of this because it would seem to me that this would cause different samples to get unequal treatment.
I was wondering if anyone else had any experience with this particular item and what you did. I have considered trying to either tape the tube holder to the edges or using a weight to hold it down.

I also use the Misonix Sonicator 3000 with the cuphorn and so far I have no problem. The size of DNA fragments have been very reproducible. There are a couple of tricks to this:
1. I use 2ml microtubes

These fit well into the tube holder with part of the cap slightly jutting out and thereby make the tube holder sits tightly in the cuphorn.
2. Make sure that the water level is not too high; the bottom of the microtubes have to touch the metal part of the sonicator
