troubleshooting in RNA extraction with Trizol - (Jul/23/2007 )

i work with MCF-7 , breast cancer cell line. i got a problem with my RNA extraction. the upper aqueos phase showed pink colour and it was turbid after adding Isopropanol. can anyone give me some suggestion?

Try to clean the aquaeous phase once more with chloroform/BCP before you advance to the precipitation step.

well i think your homogenization wasn't well done as the upper phase should'nt be pink. You have encountered phase inversion.
In such case, re homogenize and respin your homogenate.
i would suspect you didn't add enough trizol. try to double the amount if the respin still gives pink phase upper.
