Closing up restriction sites - KpnI/XhoI site needs to get closed (Jul/23/2007 )

Hi All,

I've got 2 receptors in a plasmid. I don't need the 2nd receptor and can cut it out with KpnI and XhoI. How can I then close up the linearized plasmid considering that KpnI and XhoI are not compatible? I want to keep the 1st receptor intact in the plasmid.

Is there an enzyme I can use?

digest the 2nd receptor out and purify the DNA from gel and using klenow you could make the ends blunt and then religate the plasmid.

After I posted, I thought about Klenow - just wanted to make sure I wasn't missing anything else.
