Cell culture sterility Checks (DMEM) - (Jul/23/2007 )

Hi everyone,

I have a question regarding to a new tests I had tried into my cell culture lab. The story is: Before each time I prepared complete DMEM (with 10% FBS and 1% Pen/Strep) I took 10ml into a flask to check the sterility but I only did that visually (and it's not very accuracy)
I was searching for other methods and I bought two mediums:

Tryptone soya broth: to check aerobics
[indent]
Thioglycollate medium: to test anaerobics

Both mediums are already prepared by OXOID.

I did contaminate one bottle of DMEM (without Pen/Strep) and I aliquot 1ml of that in each medium but after 4 days I don't have any difference, it looks clean (i clean a pc keyboard with a cotton stick and later I put into the DMEM bottle!!)

What I'm doing wrong?? unsure.gif

Did U try spitting or sneezing in it? :

Maybe the keyboard is simply not dirty enough. More precisely the bacteria/fungi growing there don't particularly enjoy DMEM as a medium.

That's right!

I took a sample from my mouth with a cotton stick and after a couple of hours everything was cloudy!!
