phoenix 293 retroviral infection - (Jul/23/2007 )

hi everybody,
so I have to produce retrovirus to infect some cell for the first time, for making the retrovirus we will use the phoenix 293 amphotropic cells, but i was wondering if I have to transfect them also with a packaging plasmid or they produce it by them self.

293 phenix cells doesn't need a helper plasmid such as pCLA10. With the features of a plasmid such as pBabe, they can produce retrovirus by themselves.

thank you so much so i will not use the MLV-psi packaging virus plasmid that we where using before...

normallly, psi signal are holded by the plasmid you transfect.
i've attached a simplified map of the plasmid i use.

i will use a pBABE puro plasmid that ,looking at the map, contains the psi emelent
