southern blot SSC bufffer doubt - do we need to use electrophoresis grade sodium citrate? (Jul/18/2007 )

hi all

i am planning to do Southern

since i am doing it for first time in this lab( not very rich and also not very prompt in ordering for foreigners ) and the only available Sodium citrate is Sodium citrate dihydrate from some local company

i was wondering will it do for SSC buffer?

there should be no problem, just make sure that the fw difference (assuming that your original calculation is for a different degree of hydration) is accounted for in your calculations.

the only concern you may have is the grade (purity) of the preparation.
