Swine alveolar macrophages culture and PRRSV titer - (Jun/28/2007 )
Dear all,
I am trying to inhibit PRRSV replication in experimental infected Swine alveolar macrophages (SAM) by treating SAM with antisense Oligomer.
This oligomer is completely inhibiting PRRSV replication in-vitro in African green monkey kidney cell lines and do not found virus RNA copies by Real-time PCR Or titer by TCID 50 after oligomer treatment.
But in SAM, following the same mechanism of oligomer treatment, we found no CPE development in SAM but getting virus titer in TCID50 and RNA copies in Real-time PCR similar to virus control (no oligomer treatment). But in virus control have severe CPE development.
I don’t understand why and how I am getting virus titer (SAM) without the CPE development, and the titer of the virus is equal to the titer of virus control which have severe CPE.
Any help will greatly appreciate.
