Poly-lysine treatment and cell removing - (Jun/27/2007 )

Hi,

Next time I will try polylysine treatment to improve the attachment of my cell in 24well. Iwill do this to reduce the lost of cell during some washing.
But the question is: Can I detach my cell after a polylysine treatment and can I use the same method. In my case I use PBS-EDTA to detach the cell.

Is someone can help me??

You can detach cells after poly-lysine treatment. The cells are better attached and not completely stuck to the wells.

Do you want the cells to be intact while detaching or for making protein lysates.

I need to detach my cell to realise a trypan blue or cell lysate. Does it cause any problem to the cell??

If you want to analyse the cells, try trypsin. It should help you to detach without much damage.

Or you could try citric saline. The cells are quite intact.

if you want to make cell lysate, add the SDS buffer and everything will come off the surface.
