lysis buffer- protease inhibitors - (Jun/26/2007 )
Hi all,
I don’t have much experience with apoptosis.
I should assess caspase-9 activity using a glo-type lumiscent kit.
This is the composition of the Lysis buffer that I am going to use:
20 mM Hepes-KOH, pH 7.5, 250 Mm saccharose; 50 mM KCl; 2.5 mM MgCl2; 1Mm Dithiothreitol. Does it need to be supplemented with protease inhibitors?
Any tips or suggestions will be very much appreciated.
Thanks

as caspases are itself proteases you may block the effects you like to analyze by unspecific use of protease inhibitors
