Why is an antibiotic free media necessary before transfection? - (Jun/20/2007 )

Hi,

Could anyone tell me why you cannot have pen/strep in your media prior to transfection if the antibiotic you are using for selection is neomycin (and there is no pen/strep resistance marker on the plasmid)?? Does the antibiotic interfere with the transfection process??

During transfection, the cell is forced to take up foreign materials. Too much antibiotics entering the cell may be harmful.

I do not know if it is right; we successfully transfect even in the presence of pen/strep;

I transfected cells using FUGENE in the presence of pen/strep. It worked with 80-90% efficiency.

The antibiotic doesn't always interfere but according to Fugene, it's reccomeded to transfect without antibiotics (any antibiotics) to increase efficiency. The abs can interfere by forming complexes with the transfection reagents (according to fugene)
