Hello,
can someone tell me,
can the salt caontamination effect the PCR

I don't understand your question. Do you mean you have impure salt used? (I imagine you mean MgCl2) Or your PCR had salt from somewhere else? DNA polimerases are dependent on [Mg2+], so they are an important factor.

By the way, I don't think this is the best sub-forum to ask for this kind of help... unsure.gif

If i want to do PCR is salt contamination will affected my result?

Yes, NaCl contamination will inhibit your PCR. If NaCl contamination is coming from your DNA extraction, washing the DNA pellet twice with 70% Ethanol should get rid of salts after NaCl precipitation (depending on how you are extracting the DNA of course)

You mean extra salt in PCR system? Of course it can affect the PCR result, due to reduce the activity of the DNA polymerase.

Inhibition by NaCl is specific for each DNA polymerase. For Taq, 25 mM will exhibit slight inhibition, and 50 mM will result in strong inhibition.
