3D matrigel cultures - Help, my matrigel is out of control (Jun/14/2007 )
Hi!
I am trying to set up some 3D matrigel cultures in my lab but the matrigel do not set properly. My gels are just wet and runny even after an overnight incubation at 37 degrees. What I do is that I mix the matrigel 1:1 with ice cold PBS and just put it in the plate... Does anybody have any ideas of what I could do to improve the hardness of the gel?

Is it necessary to dilute your matrigel?
I think the only one thing to do to improve the hardness of your gel is to don't dilute the matrigel or to dilute it less.

as I remember original stock from BD must be diluted 1:7; if you dilute more, it will not become hard;
an alternative, more cheap, is collagen

Another alternative is Extracel (www.glycosan.com) If you call them and tell em what you're doing they'll probably do a little wheeling and dealing with ya.
