problem in DNA extraction from glycerol-ethanol embedded tissue - (Jun/13/2007 )

Hi, everyone!

I have a problem in DNA extraction from ethanol and glycerol-ethanol embedded tissue. I have samples of very small parasites. The parasites were treated with ammonium picrate glycerine or glycerol and determined using a microscope. Then they were moved to 96% ethanol. For isolation DNA I used Qiagen Kit, tissue normal methods. But unfortunately, I got no result. Could anybody send me a protocol which works better? Do you have any experience with ammonium picrate glycerine or glycerol treated samples?

i would ask invitrogen if DNAzol is compatible with these stored tissues. If you have columns kits, something may block binding and/or elution
