pH of western blotting buffers - pH of western blotting buffers (May/29/2007 )

hi all,

why a pH of a western blotting buffer should not be adjusted ?
western blotting buffers should have pH between 8.4 and 8.6.
My buffers are never in this pH range. mad.gif

You mean PBST? TBST? that's PH7.4
Or you mean something else?

well i use a TBS-T which pH is 8....
so i guess the value should be roughly 7.5 - 8

because, if you add the correct amounts of the specified buffers (tris and glycine, specified form (eg. tris base) and hydration) then the buffer will fall in the pH range specified.

if yours are not falling in the range then you may be using the wrong form of buffer (eg. tris hcl) or your water is not good or ...
