Invassion assay - (May/26/2007 )
i am doing invasion assay of some cells lines. My problem is when i am doing with chemoattractent (either serum in upper chamber or in lower chamber) its non invasive but when i am doing with no chemoattractant is highly invasive which suppose to be non-invasive. can you give your opinion in this regards.

What system do you use to do invasion assay? Coated protein - matrigel or collagen I type or another? What time incubation do you apply to observe effect?
Sometimes serum effect on invasion ( depends on concentration) varied in wide range ( till invasion inhibition )
Try to use another chemoattractants like condition media from 3T3 fibroblasts
You should not use serum in upper chamber but only in low chamber. If you use serum in upper chamber invasion will not go!
