How to detect apoptosis in response to a chemical? - (May/21/2007 )

If i want to follow apoptosis in suspension cells in response to a chemical, (probably going to be using flow cytommetry and fluorescence microscopy and looking at GFP and PI concurrently) which is the best kit to use? I think the pathway goes through JNK and then mitochondria - is there a kit that will indicate this?

The flow cytommetry is a good and bona fine method.

There are kits available, I used a kit were annexin-v was labeled with FITC and PI.

Hi Donna,
you can use the XTT or MTT assay to check cell viability post treatment. if your very rich you can also go for ATP glow assay from promega(its very simple but easy remember it has some drawbacks).
but the best solution is doing the annexin-PI staining where you confirm the cell death is due to apoptosis or not
