slide fixation - (May/18/2007 )
dear all
i want to do some IHC without perfusion. it works great when perfused, but without perfusion sucks. does anyone have any suggestions for fixing on the slide? im using mouse brain sections cryostat cut. i've tried methanol, acetone, ethanol and 4% PFA fixation on th eslide but results aren't great.

We get some brains which are not perfused. But once in our hands, we put the brain in 4% PFA overnight at RT and then process it for paraffin embedding and section it.
You could try the same for your samples, then dehydrate them in sucrose and then try sectioning.
