macrophages with dialyzed FCS - (May/02/2007 )

Hi everybody!

I am working with BM derived macrophages and I get really nice and many cells as long as I use normal FCS. But when I use dialysed FCS (10%) my cells don't look great they are detached and I get very few cells. Does anybody have experience with dialysed FCS? What could I add to the medium? I tried B-ME but didn't help. Any suggestion?

Thanks for the link. Unfortunately I don't have access to the paper(it is too old). Could you give me advice how to replace the glutathione in the media?

why are you dialyzing your FCS? You should not have to...

why are you dialyzing your FCS? You should not have to...


I know. But in this experiment I have to. We would like to quantify cell surface proteins with MassSpec and to distinguish between treated and untreated we use heavy and light aas and the undialyzed FCS decreases the sensitivity of the approach.
Could you give me advice how to replace the glutathione in the media?
