partial digestion of Tth111I - (May/01/2007 )

have you ever tried partial digestionof Tth111I???
i want to do this but i am bit confused how to do it. the digestion temp is 65c. i cant decide the digestion time , DNA amount, enzyme amount
and also how to inactivte the enzyme

Just set a dilution series (1:3, 1:9, 1:27, etc) with the RE enzyme with your DNA and digest for a fixed time (30 minutes is a good start). To stop the enzyme add 1 vol of 50mM EDTA and phenol extract.
