same protein=opposite effects - (Apr/30/2007 )

Hi!

I'm analyzing the role of a specific protein on proliferation. I thought it'd be a good aproximation to overexpress and downregulate the protein using a plasmid and siRNA respectively.
Surprisely, I observe that when I overexpress or downregulate protein, both situations inhibit cell proliferation!! wacko.gif
So my question is; is possible that the same protein has opposite effect depending on its cellular levels?

signal transduction pathways are not linear or only very partially; we are faced with networks, and if you analyze a protein that works as an important node in a network, integrating up- and downregulations, you may find effects that seem to be diametral at first glance but reflect the special importance of this protein;

I would take a strong look at kinetics of regulation
