monocyte isolation from bone marrow - primary monocytes from murine bone marrow (Apr/25/2007 )

Hi there,

Does anybody have any experience of isolating monocytes from murine bone marrow? I have found protocols for isolating peripheral blood monocytes from mice, but I don't think this method would yield the number of cells I require. I am hoping to perform chemotaxis assays using the purified monocytes.

Any help or advice would be greatly appreciated!

Hi,
Monocytes isolation from bone marrow done by following way
Renoved leg bone from mussles
Cut both end of bone
Passing incomplete media from one end and collect from another end
then spin at 100-200 g for 5 min at 4 0 C
If you get white cells pellet then again resuspend in complete media
count them and plated in the culture plate for 2h.
After two hrs removed media only attached cells will be there again added fresh media
and now start their experiment.
