H there,

as I understand, you're about to isolate DNA from blood, and the Qiagen kit you mentioned is perfectly good for that. You can download the manual from their hompage, and it comes with the kit, too, of course....

Just to be sure that I understand your question:
- you have finished your experiment
- you are writing a report
- you want to describe, how the DNA extraction that you did works
- you have the QIAMP kit and in the kit you have the handbook, that tells you, how to do the extraction with the kit. This handbook is the manual!!!
Did I get your question?
If yes, the answer is really easy:
In the handbook, there is a section called Principle and Procedure. In this section, they explain the principle behind the DNA extraction (in fact, it is a silica based column and buffer system, which specifically binds DNA).

And, pardon that I start to quote myself ... size doesn't matter biggrin.gif tongue.gif biggrin.gif

Ah, good point there, kruemelmonster, I think you got him(or her) right... And for the size thing:

I'd say, size can make a difference rolleyes.gif

basically, dna extraction kits came after the alkaline methods developped in 60's-70's
