About the adenovirus infection efficiency on tissue culture - (Apr/01/2007 )

Hi,
Recently I am using an andeovirus to silence a gene in the fetal lung tissue culture, the tissue are from day 19 fetal rat lung, and cutted into a 1mm3 small slice. But I found the silence efficiency is very low. And when I am checking the virus infection on tissue section, I found only the surface cells are infected. Do you know how can I increase the infection efficiency on lung tissue?

There is not much you can do about it. Adv is 100 nm in size and it needs to penetrate into the tissue in order to transfect cells that are not exposed. Maybe you can digest your tissue samples into cells and get better transfection this way.
