too many dead cells why? - (Apr/01/2007 )
goodmorning everyone
i have a problem with my 293cells *its stably transfected with 2 genes* i always find dead cells floating even next day of passage?
one friend said may be its because of long trypsinization but i only leave it for one minute or so in room temperature?
my starting cells confluency*the one i do passage from* sometimes be 90% confluent and sometimes 70% sometimes 100%?
so may be this is the cause?
i change media between passage by the way?

maybe you did pipeting little too much and cell died because of that?

i am doing passage for 2 other cell lines by the exact same way , one is 293A parent cells and the 293 stably expressing one gene only and i only have this problem with 293 stably expressing 2 genes so is there any correlation?

transfections may alter properties of cells, f.i. faster or slower proliferation, tighther or loser attachment, more or less spontaneous apoptosis etc.
it strongly depends on your transfected genes and their expresssion, respectively
