GST / MBP tags - (Mar/12/2007 )

Hi,

has anybody experiences with the GST and MPB "tags" in Staphyolocci? His/Arg/Strep-tags are quite nice but those other tags are working very well for "difficult" proteins.

Hope you have some good advices!

Not worked in staph but have made several proteins tagged to MBP. Advice.... just because your plasmid is recombinant, does not mean it will express protein... even if you have sequenced it and it is correct. Adding glucose to the media is critical. If you can, clone BamHI to HindIII. My best expressors are that say. Omit the hydroxyapetite steps. Elute and dialyze instead.
