why is my compound crystallizing out of the media - in a 72hr cytoxicity assay, the compound being tested crystallizes out (Mar/06/2007 )

hi, I need your help....

I am trying to test the anticancer activity of several pure compounds which I have dissolved in DMSO.
For the most part the compounds are soluble DMSO however there are some that ar not readily soluble and require substantial vortexing and in some cases slight heating.
After dissolving and application to media in wells containing cells, the assay is incubated for 72 hrs. What I have observed is that, even with compounds that fully dissolve in DMSO, between 48 and 72 hrs at 50 and 100uM, crystals of the compound are forming. What can be done? Is there an alternative solvent other than DMSO....I have not seen any publications that have utilized methanol, my assumption is that it is too toxic.....

am I right? your stock solutions crystalize? Not the compound added to cell culture?

you may presolve in DMSO and then add some water or a buffer to a defined conc

alternatives to DMSO are DMF, methanol, ethanol, isopropanol

your compounds crystalize cause the solvent evaporates. you can either decrease its concentration or seal the cells very very well.

Hi,

well I am not referring to the to the stock solutions crystallizing out, but rather after adding the compounds to the wells in the plate, the cpmd is crystallizing out...
