MACROPHAGE ADHESION TO TEST TUBES? - (Mar/06/2007 )

hi,
i'm having problems with my THP1 derived macrophages. I used PMA (50ng/ml) to differentiate my THP1 cells into macrophaes, and when i scrape them and put them in a centriguge tube they keep sticking to the plastic (no matter how many G-s i use). mellow.gif
i cannot use trypsin or EDTA becuse of the nature of my experiment...
I've also tried to centrifuge in glass tubes, but nothing seems to help!
anybody got an idea?

did you use polypropylene tubes for centrifugation?

i agree ! polypropylene tubes support less adherence. also if the problem persists try keeping the tubes at 4C.. on ice. lower the temperatures lower is the adhesion

We grow our murine macrophages in suspension because of this adherence problem. Try and use Borosilicate glass. This again is a product which does not allow the cells to adhere.
