FITC and PE Overlapping emissions in Flow Cytometry - (Feb/13/2007 )

I have ordered antibodies conjugated to either FITC or PE and am concerned that there will be an overlap in emission. If so, how would I compensate for this overlap?

yes, there will be overlap...probably by about 25% but you should run single stains to see exactly how much

go to omega optical "curv o matic" website to see graphs of fluorochrome emission and excitation.
do your antibodies need to be pre conjugated could you not simply label your own thus choosing the wavelenghs you use
