NK92 cell line - (Feb/08/2007 )

Hi,

Does anybody have experience with natural killer 92 (NK92) cell line, the problem we are experiencing is that the cells use all the glucose from the media and don't look healthy especially after week-end. Cells are maintained in Alpha MEM (Gibco) supplemented with 12.5% FBS and horse serum, 2.5mM L-Glutamine, 100µM B-ME and 200 IU IL-2 (PeProTech).

I think I am using the similar medium as you. Healthy NK92 should form clumps and even very big clumps. How do your cells look? I have hydrocortisone in my medium with the final concentration of 10-6 M. It's a nonspecific cell-growth stimulator. You can try.

Thank you for your reply, I'll try.
Our cells look fine with lots of clumps, big as well as smaller one, but the issue is that we do bioassay (measure release of IFN-gamma in the media) using these cells and 2 days after seeding for the assay the glucose level drops down to zero when we measure metabolites with "bioprofile 400". I guess when glucose level is zero the cells are stressed which might influence the results?
